What is Ipamorelin?

Ipamorelin is a synthetic five-amino-acid peptide supplied for in-vitro laboratory research. It includes two non-standard amino acids: Aib (α-aminoisobutyric acid) and D-2-Nal (3-(2-naphthyl)-D-alanine) [1].

It was first described in 1998 as a growth hormone secretagogue: a compound that acts at the growth hormone secretagogue receptor (GHSR), also called the ghrelin receptor [1][4]. GHSR is a G protein-coupled receptor.

What has been studied? (preclinical receptor research only)

• Original pharmacology. The first paper describes how Ipamorelin came out of a chemistry programme based on GHRP-1. In primary rat pituitary cells it released growth hormone with an EC50 of 1.3 ± 0.4 nmol/l, similar to GHRP-6. Work with receptor antagonists showed it acts through a GHRP-like receptor [1].
• Receptor structure. Cryo-EM structures of GHSR bound to ghrelin and to a synthetic agonist show the binding pocket, and point to a salt bridge and an aromatic cluster as important for receptor activation [2].
• The ghrelin receptor pocket. A crystal structure of GHSR bound to an antagonist shows a binding pocket split by a salt bridge (E124–R283), and a wide hydrophobic gap between two of the receptor’s helices [3].
• Receptor signalling. A review sets out GHSR’s molecular structure, the signalling pathways it can trigger in different cell types, and how the receptor is regulated [4].
