What is Semaglutide?

Semaglutide is a synthetic analogue of the peptide hormone GLP-1, supplied for in-vitro laboratory research. It acts at the GLP-1 receptor (GLP-1R), a class B1 G protein-coupled receptor [1][2].

What has been studied? (preclinical receptor research only)

• Molecular design. The discovery paper describes how the fatty-acid group and its linker were chosen to raise binding to the blood protein albumin and to resist enzyme breakdown, while keeping GLP-1R potency. It reports a GLP-1R affinity of 0.38 ± 0.06 nM [1].
• Chemistry review. A review sets out the chemistry used to design long-acting GLP-1 analogues, including the fatty-acid and linker combinations studied for albumin binding [3].
• Receptor structure. Cryo-electron microscopy (cryo-EM) structures of Semaglutide bound to the GLP-1R–Gs protein complex show peptide contacts similar to native GLP-1, with different motions in the receptor and peptide [2].
• How peptides switch on GLP-1R. Structural and functional studies of peptide agonists suggest that receptor flexibility, linked to the peptide’s N-terminal region, helps decide how strongly a peptide activates GLP-1R [4].
