What is Tirzepatide?

Tirzepatide is a synthetic peptide supplied for in-vitro laboratory research. It acts at two related receptors: the GIP receptor (GIPR) and the GLP-1 receptor (GLP-1R) [1][2]. Both belong to the class B1 family of G protein-coupled receptors.

What has been studied? (preclinical receptor research only)

• Signalling at each receptor. Cell studies found that Tirzepatide behaves like native GIP at GIPR. At GLP-1R it favours cAMP generation over β-arrestin recruitment, and drives less receptor internalisation than GLP-1. The authors describe this as “imbalanced and biased” agonism [1].
• Signalling pathways in HEK293 cells. A separate cell study reported that Tirzepatide (tested as LY3298176) is biased towards ERK1/2 phosphorylation relative to cAMP at both GIPR and GLP-1R, compared with each receptor’s own hormone [4].
• Receptor structures. Cryo-electron microscopy (cryo-EM) and molecular dynamics studies show how Tirzepatide binds GIPR and GLP-1R. They report that the fatty-acid modification, together with the amino-acid sequence, shapes how it engages each receptor [2][3].
